ABSTRACT

@#AIM: To investigate the effect of inclination and eccentricity of intraocular lens(IOL),on optical imaging quality <i>via</i> wavefront aberration optical path system.<p>METHODS: The spherical IOL Sensar AR40e, the aspherical monofocal IOL Tecnis ZA9003, and the aspheric multifocal IOL Tecnis ZM900 were measured at the center of the center using a laboratory-built Hartmann-Shack IOL wavefront aberration path system at 5.0mm simulated pupil diameter. 0, 0.2, 0.4, 0.6, 0.8mm, the effect on the optical imaging quality when tilting 5°, 10°, 15°, 20°, 25° to the nasal side and the temporal side, and quantitative imaging quality by high-order aberration and modulation transfer function.<p>RESULTS: The Temnis ZA9003 MTF value was higher than AR40e and Tecnis ZM900 when inclinationed within 5°, while the three IOL MTF values were significantly different when inclinationed 5°, 10°, 15°, and 20°. Tecnis ZA9003 The IOL inclination angle was significantly positively correlated with the coma(<i>r</i>=0.842, <i>P</i><0.001), and there was no significant correlation with the spherical aberration(<i>r</i>=0.229, <i>P</i>=0.241). The three IOL MTF values were obtained when the eccentricity was 0.6 and 0.8 mm. Significant differences(both <i>P</i><0.001), the imaging quality of the Tecnis ZM900 eccentricity greater than 0.4mm decreased significantly.<p>CONCLUSION: When the inclination of aspheric IOL(-0.27μm spherical aberration)is less than 5°and the eccentricity is less than 0.4mm, aspherical IOL has a better imaging quality than that of spherical IOL. Tecnis ZM900 IOL has a lower optical imaging quality than that of spherical and aspheric IOL when the eccentricity of IOL is more than 0.4mm.

ABSTRACT

OBJECTIVE: To explore the effects of occupational noise exposure on the levels of serum homocysteine( Hcy)and blood lipid in workers. METHODS: By cluster sampling method,247 workers from 3 noisy operational factories in Shandong Province were selected as exposure group,and 198 non-noise exposed workers from the same factories were chosen as the control group. The levels of Hcy,triglyceride( TG),total cholesterol( TC),high-density lipoprotein( HDL) and low-density lipoprotein( LDL) were examined using enzymatic cycling assay. The influence factors of high Hcy levels in workers were analyzed by multiple Logistic regression analysis. RESULTS: The over standard rate of 8-hour equivalent sound level in workplaces of the 3 factories was 44. 4%-50. 0%. The levels of Hcy in male and female workers of exposure group were higher than those of the same gender workers in the control group( P < 0. 01). The levels of HDL in female workers of exposure group were lower than those of the female workers in control group( P < 0. 05). There was no statistical significances in the levels of TG,TC and LDL in the control group and exposure group( P > 0. 05). The detection rate of high Hcy in exposure group was higher than that of the control group( 91. 50% vs 56. 57%,P < 0. 01).Noise exposure and gender were the independent influence factors causing high Hcy levels in workers( P < 0. 05). The odds ratio( OR) of high Hcy in noise exposure workers was 2. 78 times of that of those non-exposed workers. The OR of high Hcy in male exposed workers was 6. 21 times of that of female workers. CONCLUSION: Occupational noise exposure could lead to the increasd Hcy levels in workers.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the radiosensitizing effect of nitric oxide (NO) combined with radiation on esophageal cancer cell line TE-1.</p><p><b>METHODS</b>Methyl thiazolyl tetrazolium (MTT) assay was used to assess the effects of NO and radiation on TE-1 cells regarding inhibition of cell proliferation. Flow cytometry was used to examine the effect of NO and radiation on cell apoptosis and cycle. Reverse transcription polymerase chine reaction and Western blot were used to evaluete the effect of NO on mRNA and protein expression of manganese superoxide dismutase (MnSOD).</p><p><b>RESULTS</b>NO inhibited the proliferation of TE-1 cells while significantly enhancing their radiosensitivity. The application of NO combined with radiation significantly increased the apoptosis rate and G2/M phase proportion of TE-1 cells, with substantial decreases in the MnSOD mRNA and protein expression levels.</p><p><b>CONCLUSIONS</b>NO reduces the MnSOD mRNA and protein expression levels by affecting TE-1 cell cycle, further inhibiting the apoptosis of esophageal cancer cells and enhancing the killing effect of radiation on esophageal cancer cells.</p>

ABSTRACT

In term of four aspects, named "getting the familiarity", "turning to the positive action", "conquering the fear emotion" and "bringing the practice into play", the authors state that the teaching pattern, "training the techniques of acupuncture and moxibustion before introducing meridians and acupoints" can vividly introduce the classical content of meridians and acupoints in the Part One of Science of Acupuncture and Moxibustion, exclude the repeatability of the content and improve the teaching quality. This teaching pattern enables the students mastering the techniques of acupuncture and moxibustion before the practice and laying a good foundation for learning meridians and acupoints. The authors put forward the good suggestion of the teaching course arrangement of Science of Acupuncture and Moxibustion to benefit the teaching reform.

ABSTRACT

<p><b>OBJECTIVE</b>To construct a recombinant lentiviral vector for manganese superoxide dismutase (MnSOD) gene expression, and observe its effect on the proliferation of esophageal cancer cells in vitro.</p><p><b>METHODS</b>Chemical methods were employed for synthesis of the MnSOD cDNA sequence sections, along with the attB sites. Target gene fragment was constructed on the pMD-18T vector, and the recombinant plasmid pDONR221 was obtained after BP recombination reaction. Sequencing was followed by LR recombination reaction between the plasmid and DEST to obtain the lentiviral vector, which worked with helper plasmid for co-transfection of human embryonic kidney epithelial cells (293T cells). Amplification was done to determine its titer, and both transfection and selection procedures were made to get two stable transfected esophageal cancer TE-1 cell lines with medium MnSOD expression (TE-1Mm cells) and high MnSOD expression (TE-1Mh cell), and empty vector cell (TE-1Mn cells). Reverse transcription polymerase chine reaction (RT-PCR), immunofluorescence, immunocytochemistry and Western blot were used to detect the target gene with respect to its expression in the TE-1 cells. Additionally, colorimetric 3-[4,5-dimethy thiazol-2-yl]-2,5-diphenyltetrazolium bromide (MTT) assay, agar colony formation assay, annexin V-FITC/PI staining and flow cytometry experiments were also conducted as to observe the influence of the medium and high MnSOD overexpressions on the proliferation of esophageal cancer cells.</p><p><b>RESULTS</b>RT-PCR indicated that the transfected TE-1 cells showed positive MnSOD expression at different levels. Immunofluorescence, immunocytochemistry and Western blot suggested that TE-1Mm cells and TE-1Mh cells had MnSOD protein expression at different levels. MTT assay indicated that TE-1Mm cells had a significantly decreased survival rate compared with that of the two control cells (TE-1 cells and TE-1Mn cells), and TE-1 Mh cells had an significantly increased survival rate (P<0.05). The colony formation ability of TE-1Mm cells was (23.0 +/- 2.7)%, and that of TE-1Mh cells was (45.3 +/- 4.5)%, significantly different form the (34.7 +/- 4.2)% in TE-1 cells and (33.7 +/- 4.7)% in TE-1Mn cells (P<0.05). Annexin V-FITC/PI double staining experiment of the stably transfected cells cultured for 48 h showed that the early apoptosis rate in TE-1Mm cells was (10.6 +/- 1.0)%, significantly higher than (2.6 +/- 0.2)% in the TE-1 cells, (2.5 +/- 0.6)% in the empty vector cells and (1.0 +/- 0.1)% in the TE-1Mh cels (P<0.05). The fluorescence index (FI) of mitochondrial apoptosis of TE-1Mm cells was 0.948 +/- 0.019, significantly lower than that of TE-1 cell (1.000 +/- 0.022) and empty vector The fluorescence index of TE-1Mn cells (0.997 +/- 0.023) and TE-1 cells (1.000 +/- 0.022) were significant different from that of 0.948 +/- 0.019 in TE-1Mm cells and 1.076 +/- 0.022 in TE-1Mh cells, indicating a significant difference of mitochondrial apoptosis between the cell groups. FCM results indicated that the ROS fluorescence index of TE-1Mm cells was 0.859 +/- 0.040, that of TE-1Mh cells was 0.763 +/- 0.039, significantly lower than that of TE-1 cells (1.000 +/- 0. 042) and empty vector cells (1.002 +/- 0.047) (P<0.05).</p><p><b>CONCLUSIONS</b>Stably transfected cell lines with MnSOD expression have been successfully established. MnSOD overexpression shows bidirectional effect on the proliferation of esophageal cancer cells.</p>

ABSTRACT

To try to find out the possible use of histochemistry in the prenatal diagnosis in first trimester of pregnancy, 20 kinds of enzymes in chorionic villi from 6th to 9th week of gestation which are related to inborn errors of metabolism were investigated and graded according to their histochemical reactive intensity; the enzymes listed below: (1). ?-glucuronidase, (2). ?-galactosidase, (3). arylsulfatase, (4). ?-N-acetylglucosaminidase, (5). 3?-hydroxysteroid dehydrogenase, (6). NADH dehydrogenase, (7). adenosine triphosphatase, (8). alkaline phosphatase,(9). acid phosphatase, (10). creatine phosphokinase, (11). glutamic dehydrogenase, (12). aldolase, (13). xylitol dehydrogenase, (14). alcohol dehydrogenase, (15). xanthine oxidase, (16). catalase, (17). ornithine carbamoyl transferase, (18). lipase, (19). cholinesterase, (20). ?-glutamyl transpeptidase. The experimental results show that the last 6 kinds of enzymes present negative reactions. Since the his- tochemical reactions of the first 14 kinds of enzymes are positive, it is possible that they can be used for the prenatal diagnosis of inborn errors of metabolism caused by dificiency of corresponding enzymes. The results also show that compared with biochemical method, histochemical method has the advantages of requiring smaller amount of chorionic villi, being without contamination by maternal cells, simplicity and rapidity.
